Dissertation Title: Evaluation of genetic polymorphism of in selected broodstocks of paddlefish (Polyodon spathula Walbaum 1849) based on microsatellite DNA sequences

2007-present   Employed in Department of Environmental Biotechnology

Teaching activity

genetic, ecotoxycology, and some other subjects including aquaculture, molecular and populaton genetic topics

Research Interests

Aplication of the microsatellite markers in:

1) Estimation of genetic variations within domesticated stock of commercial important species (paddlefish), endangered populations of salmonids, sturgeons and small cyprinid species (lake minnow) and genetic distance between them

2)  Management of the genetic variation resources within commercial stocks and natural populations

3) Enrichment of the genetic variation, and preventing of the inbreeding events within commercial stocks by assemblage pairs of spawners pairs following to their individual genetic differences (microsatellite loci)

4. The verivication of gynogenesis and androgenesis results applying sets of  microsatellite markers

The list of most important methods and techniques that I apply during my studies

Field based techniques:

  • Fish marking (microchip markers)
  • Tissue sampling

Molecular techniques:

  • Extraction of DNA from various tissues (fragments of fins, skin, muscles)
  • Amplification of microsatellite loci using PCR technique (primers designing, optimisation of reaction parameters)
  • Assemblage of primers sets for multiplex PCR reactions
  • Measurement of DNA fragments length using automated capillary DNA sequencers (Beckmann Coulter, Applied Biosystems)
  • Genotyping of microssatelite loci using  automatic capillary sequencers

Mathematical and statistical methods:

  • Estimation of genetic variation within populations and differences between them, evaluating of bottleneck events in population’s history,
  • Establishing of individual genetic profiles of fish,
  • Arrangement pairs of spawners in order to increase the heterozygosity of their progeny
